However, phenylphosphorothiamine must be dephosphorylated by alkaline phosphatase to become a lipid soluble precursor before it can penetrate the cell membrane and exert biological effects.

Compared with other derivatives of thiamine, Benfotiamine CAS 22457-89-2 is absorbed more quickly and completely after oral administration. Phenylthiophene is rapidly metabolized into thiamine after absorption into the systemic circulation, without increasing drug concentration in the brain. Benfotiamine CAS 22457-89-2 also generates hippuric acid and is excreted through urine.
